I've been given an scanjet 2400 by work, and discarded the bundled
software as it won't save to disk in jpeg format. I can run it from
PSP using the bundled twain driver, but every time I invoke it I have
to reselect my preferred resolution and colour settings (256 grey,
300dpi as it happens).

I can't find any way to set these as a default. There is a
preferences menu item, but it is only about dross like lamp life and
preview modes, nothing to select preferred settings.

Does anyone know how to do this?

It should save the last settings used.
Look for a Read Only .ini file for the scanner. Reset the Read only if
found.
One scanner is had could save a setting profile and you just loaded the
profile you wanted.
