Okay, this is going to be a very easy question for someone, it just
isn't easy for me.

I just profiled my Canon FS4000 scanner using a target slide and
Vuescan. Okay, it was really easy (almost too easy). I scanned a
couple of reference slides and see a very, very slight difference. I
tried playing with curves to get a match between old (unprofiled) and
new (profiled) and found the task to be very hard.

Here's the question, how do I 100% convince myself that there is a new
profile being applied? If I hadn't profiled correctly, would Vuecan
apply some predetermined profile when I use ICC profile?

I was thinking about simply scanning the it8 target again and use the
built in versus profile and check a few blocks on the scan. Is that
the best way?

Well, here's a quick and easy way to check.
In Vuescan, go to the color settings, set color balance to neutral and
toggle between scanner ICC profile "built-in" and your new profile
(vuescan.icc?) for a few scans where you know what the slide should
look like.

You can also copy the new scanner ICC profile to
windows/system32/spool/drivers/color (I think- somewhere around there
you'll see a lot of .icc profiles if you're using WinXP).
Next, make a raw 16 bit scan of a slide in Vuescan. Open it in
Photoshop and go to mode/assign profile. Switch between the Canon
FS4000 default profile and the new profile (probably Vuescan default
profile) and see if it looks any different. I've found Vuescan doesn't
correctly apply the factory profile to slides, but does a good job
applying its own profile.

was quite easy to see in the grays.
